Cytotoxicity and Selectivity Profiling

Cytotoxicity & Selectivity

Differentiate true anti-inflammatory activity from general cytotoxicity and assess selectivity across related targets to prioritize leads.

  • Multiple tumor cell lines and primary cells available for assay
  • ATP-based homogeneous viability readout for robust IC50 determination
  • Cell-dependent cytotoxicity assays with immune cell effectors
  • Selectivity index calculation across related enzymes or isoforms
  • Custom assay development and project-scoped profiling

Why Cytotoxicity & Selectivity Profiling Matters

Advancing a compound from discovery to clinical development requires accurate data on both potency and safety. Our in vitro profiling service delivers the unbiased information you need to make go/no-go decisions with confidence.

  • Distinguish target-mediated killing from off-target toxicity
  • Rank compounds by selectivity index to prioritize leads
  • De-risk candidate selection before in vivo studies
  • Identify cytotoxic vs. cytostatic effects with kinetic readouts

Our Approach

Integrated Cytotoxicity & Selectivity Workflow

We combine validated cell-based assays with rigorous data analysis to provide a clear picture of compound cytotoxicity and selectivity, tailored to your project needs.

Cell-Based Cytotoxicity Assays

We deploy ATP-dependent homogeneous assays (e.g., CellTiter-Glo) across a panel of tumor cell lines and primary cells, measuring viability with high linearity from 100 to 200,000 cells per well in 96-well plates.

  • Choose from 30+ tumor cell lines covering major cancer types
  • Primary cells including PBMCs, iPSC-derived cells available
  • Genetically engineered cells overexpressing targets on request

Selectivity Profiling Across Targets

To assess therapeutic window, we evaluate compound activity against related enzymes (e.g., COX-1 vs. COX-2) or off-target kinases using enzymatic or cell-based assays, calculating selectivity indices.

  • Enzymatic assays for isoform-specific selectivity
  • Cell-based reporter assays for pathway selectivity
  • Selectivity index (SI) computed as IC50 toxic / IC50 target

Quality Control

Each assay includes positive controls (e.g., staurosporine for cytotoxicity) and replicate wells to ensure Z' factor >0.5; dose-response curves are fitted with four-parameter logistic models.

Statistical Rigor

IC50 values are reported with performance scoped per project confidence intervals; selectivity ratios are calculated with propagated error, and batch effects are minimized by plate-level normalization.

Biological Interpretation

We provide a summary report contextualizing compound activity vs. toxicology databases, helping you identify liabilities early and refine chemical series.

Cell line panelWe offer a panel of 1–30 tumor lines plus primary cells, and custom engineering is available upon request.
Assay readoutATP-based viability (CellTiter-Glo, CellTiter-Fluor), luciferase reporter, or LDH release
Selectivity targets2–10 related enzymes/isoforms; kinase panel targets
Concentration rangeTypically 10-point 3-fold dilution series; range defined with customer
ReplicatesTriplicate or quadruplicate; with plate-level normalization

Workflow

How It Works

A streamlined process from compound submission to actionable data, designed to fit your project timeline.

Project Scoping

We discuss your compound series, target biology, and desired selectivity endpoints. Together we define cell lines, assay format, concentration range, and positive controls.

Assay Development & QC

Cell lines are cultured and verified; assay conditions optimized for signal-to-background, linear range, and reproducibility. A pilot run with controls confirms Z' factor >0.5.

Compound Treatment

Compounds are prepared at specified concentrations in DMSO (final ≤ performance scoped per project). Cells are treated in 96- or 384-well plates and incubated under optimized conditions before viability measurement.

Data Analysis

Raw luminescence is normalized to vehicle control. IC50 values are calculated using four-parameter logistic regression. Selectivity indices are computed as IC50 toxic / IC50 target.

Reporting & Review

A comprehensive report including dose-response curves, IC50 tables, selectivity heatmaps, and QC metrics is delivered. We offer a follow-up teleconference to discuss results.

Applications

FAQs

Frequently Asked Questions

Common questions about our cytotoxicity and selectivity profiling service.

  • We maintain a panel of over 30 tumor cell lines covering lung, breast, ovarian, pancreatic, colorectal carcinomas, leukemias, and lymphomas. Primary cells such as PBMCs and iPSC-derived cells, as well as custom genetically engineered lines, are available on request.

  • Yes. For antibody-drug conjugates, monoclonal antibodies, and bispecific engagers, we offer cell-dependent cytotoxicity assays using PBMCs, cytotoxic T cells, or NK cells as effector populations. We also support custom target cell lines expressing your antigen.

  • Selectivity index is defined as the ratio of the toxic concentration (IC50 in a relevant normal cell line or against an off-target enzyme) to the effective bioactive concentration (IC50 in the target cell line or against the primary target). An SI >10 is often considered desirable for further development.

  • Each report includes raw data and plate maps, dose-response curves with IC50 values and performance scoped per project confidence intervals, selectivity heatmaps, QC metrics (Z' factor, replicate concordance), and a written summary with go/no-go recommendations.

  • Yes. We can design custom assay protocols to test compounds in combination with immune effector molecules, other drugs, or in co-culture systems. Project scoping will define the appropriate conditions.

  • The expected timeline will be defined during project scoping and included in your study proposal.
